Regulation and safety
Kisspeptin-10 occupies an awkward position. It is a natural human peptide fragment and has been given to volunteers in research settings, yet it has never been taken through the development programme that leads to a licensed medicine. Its legal treatment therefore depends on how and where a product is supplied.
Medicine status
We found no marketing authorisation for any product containing kisspeptin-10 from the US Food and Drug Administration, the European Medicines Agency or other national regulators. Longer-acting synthetic analogues such as TAK-448 have been made for research and early clinical testing, but none has reached approval.
United States: compounding
On 29 September 2023 the FDA placed kisspeptin-10 in category 2 of its interim policy for bulk drug substances nominated for compounding under section 503A, the group of substances that may present significant safety risks. The agency's stated reasons were:
- possible immunogenicity, meaning an unwanted immune response, for certain routes of administration;
- complexities around peptide-related impurities and characterisation of the active ingredient;
- no, or only limited, safety information for the proposed routes, so that the agency could not tell whether the substance would cause harm in humans.
Sport
Since the 2024 edition, the World Anti-Doping Agency's Prohibited List names “kisspeptin and its agonist analogues” in section S2, under peptides that stimulate testosterone production in males. Substances in this section are prohibited both in and out of competition, and the entry remains in the 2026 List.
Open safety questions
The published human studies were not designed to assess safety beyond the short term. They were small, brief and closely supervised, and they reported hormone measurements rather than systematic safety outcomes. Several biological points remain open:
- Receptor desensitisation: continuous exposure switched off the hormone response in monkeys within hours and in men over five days. Under some conditions the same peptide can therefore suppress rather than stimulate the axis.
- Effects outside the brain: the receptor is also present in blood vessels, and kisspeptins constricted isolated human arteries in the laboratory. What this means in a living person is not known.
- Cancer biology: the KISS1 system was discovered through its effects on metastasis, and its role in different tumours is still being studied. Researchers do not regard it as fully understood.
- Unstudied groups: there are no data on pregnancy, on long-term use, or on people with heart, kidney or hormone-dependent conditions.
- Impurities and immunogenicity: these are the concerns the FDA itself raised, and they apply especially to material of unknown origin.
Why research-grade material is different
Peptide sold as a laboratory reagent is made to a chemical specification for experiments. It is not produced under the controls required for medicines, it is not tested for sterility or endotoxins in the same way, and its labelled content may not match what is in the vial.
To sum up
Kisspeptin-10 is a research peptide without approval as a medicine, flagged by the FDA for compounding and prohibited in sport. Anyone with a health concern related to it should consult a doctor.
